چکیده

This study addresses the role of store-operated Ca influx in the regulation of exocytosis in inflammatory cells. In HL-60 granulocytes, which do not possess voltage-operated Ca channels, the chemotactic peptide fMet-Leu-Phe (fMLP) was able to stimulate store-operated Ca influx and to trigger exocytosis of primary granules. An efficient triggering of exocytosis by fMLP required the presence of extracellular Ca and was inhibited by blockers of store-operated Ca influx. However, receptor-independent activation of store-operated Ca influx through thapsigargin did not trigger exocytosis. fMLP was unable to stimulate exocytosis in the absence of cytosolic free Ca concentration [Ca]c elevations. However, a second signal generated by fMLP synergized with store-operated Ca influx to trigger exocytosis and led to a left shift of the exocytosis/[Ca]c relationship in ionomycin-stimulated cells. The synergistic fMLP-generated signaling cascade was long-lasting, involved a pertussis toxin-sensitive G protein and a phosphatidylinositol 3-kinase. In summary, store-operated Ca influx is crucial for the efficient triggering of exocytosis in HL-60 granulocytes, but, as opposed to Ca influx through voltage-operated Ca channels in neurons, it is not a sufficient stimulus by itself and requires synergistic receptor-generated signals.

Refilling of cortical calcium stores in Paramecium cells: in situ analysis in correlation with store-operated calcium influx.

This is the first thorough study of refilling of a cortical calcium store in a secretory cell after stimulation in which we combined widely different methodologies. Stimulation of dense-core vesicle ("trichocysts") exocytosis in Paramecium involves a Ca(2+) -influx" superimposed to Ca(2+) -release from cortical stores ("alveolar sacs" (ASs)). In quenched-flow experiments, membrane fusion freque...

Two Modes of Secretion in Pancreatic Acinar Cells Involvement of Phosphatidylinositol 3-Kinase and Regulation by Capacitative Ca2+ Entry

In pancreatic acinar cells, muscarinic agonists stimulate both the release of Ca(2+) from intracellular stores and the influx of extracellular Ca(2+). The part played by Ca(2+) released from intracellular stores in the regulation of secretion is well established; however, the role of Ca(2+) influx in exocytosis is unclear. Recently, we observed that supramaximal concentrations of acetylcholine ...

Store-operated calcium channels.

In electrically nonexcitable cells, Ca(2+) influx is essential for regulating a host of kinetically distinct processes involving exocytosis, enzyme control, gene regulation, cell growth and proliferation, and apoptosis. The major Ca(2+) entry pathway in these cells is the store-operated one, in which the emptying of intracellular Ca(2+) stores activates Ca(2+) influx (store-operated Ca(2+) entr...

Opposing effects of protein kinase A and C on capacitative calcium entry into HL-60 promyelocytes.

Treatment of HL-60 cells with thapsigargin, a microsomal Ca2+/ATPase inhibitor, led to depletion of intracellular calcium stores followed by capacitative calcium entry. Stimulation of adenylyl cyclase with forskolin enhanced thapsigargin-induced Ca2+ influx. The forskolin effect was confirmed by enhanced fluorescence quenching induced by Mn2+ entry into fura-2 loaded cells. 1,9-Dideoxy-forskoli...
